How MOTS-c Is Dosed

MOTS-c is a mitochondrial-derived peptide, a short peptide encoded within mitochondrial DNA that is studied for its role in metabolic regulation.

There is no published human pharmacokinetic data for injected MOTS-c. The only human trial ran on a different MOTS-c analog, not the peptide itself, so its data does not transfer and we do not present a half-life curve for MOTS-c in the app.

Unlike most research peptides that are dosed in micrograms, MOTS-c community protocols are dosed in milligrams, commonly around 2.5 to 10 mg per subcutaneous injection, two to three times per week, usually from a 10 mg vial. The calculator converts your chosen dose into exact units.

The Math Behind Peptide Reconstitution

Concentration formula:

Concentration (mg/mL) = Peptide (mg) ÷ Water (mL)

Units to draw:

Units = [Dose (mcg) ÷ 1000] ÷ Concentration × 100

Example: BPC-157 250mcg dose

  • 5mg vial + 2mL BAC water = 2.5 mg/mL
  • 250mcg = 0.25mg
  • 0.25mg ÷ 2.5 mg/mL = 0.10mL
  • 0.10mL × 100 = 10 units on insulin syringe

Frequently Asked Questions

Why is MOTS-c dosed in milligrams instead of micrograms?
Community MOTS-c protocols use milligram amounts (commonly 2.5 to 10 mg per injection), which is much larger than the microgram doses of most research peptides. Make sure your calculator dose is entered correctly, since our field uses micrograms (5 mg equals 5000 mcg).
What is the half-life of MOTS-c?
There is no published human pharmacokinetic data for injected MOTS-c, so a reliable half-life cannot be stated. The only human trial used a different MOTS-c analog, not the peptide itself.
Has MOTS-c been studied in humans?
Directly injected MOTS-c has not completed published human pharmacokinetic studies. Most of the evidence is preclinical, and a separate analog has been the subject of early clinical work.
What is bacteriostatic water (BAC water)?
Bacteriostatic water is sterile water containing 0.9% benzyl alcohol as a preservative. It prevents bacterial growth, allowing multi-use of reconstituted peptides for 4-6 weeks when refrigerated.
How much water should I add to my vial?
Common choices are 1mL, 2mL, or 3mL. More water makes small doses easier to measure accurately but means larger injection volumes. 2mL is a good balance for most peptides.
What does "units" mean on an insulin syringe?
Insulin syringes use "units" as measurements. Standard U-100 syringes have 100 units per 1mL, so 10 units = 0.10mL and 50 units = 0.50mL.
How long do reconstituted peptides last?
With bacteriostatic water, most peptides remain stable for 4-6 weeks when refrigerated at 2-8 degrees Celsius. With sterile water (no preservative), use within 24-48 hours.
